Antibodies are commonly conjugated to fluorescent dyes such as FITC and Texas Red. These antibodies are widely used in research to detect cell markers. The ability of labelled antibodies to fluoresce allows proteins of interest to be localized in immunofluorescence microscopy or detected in solution by e.g. flow cytometry .

Another common modification of monoclonal antibodies is biotinylation. Biotin binds with high affinity and avidity to avidin and it’s bacterial counterpart, streptavidin. This interaction is exploited in a number of immunochemical assays, with labelled avidin/streptavidin being used to detect biotinylated antibodies bound to proteins of interest. The high affinity and low reversibility interaction permits efficient antibody detection and also the possibility of layering complexes for amplified sensitivity. Antibodies are typically biotinylated by mixing with a succinimidyl ester of biotin, which reacts with amines of lysine residues or the amino terminus of the antibody.
